Abstract: ABSTRACT: The present invention discloses a novel biofilm matrix (EPS)-responsive colour-changing paper point designed for the detection of structured microbial biofilms in root canal systems. It utilizes Concanavalin A (ConA), a lectin with high affinity for a-D- glucose and a-Dmannose residues, conjugated with a chromogenic substrate, 3,3',5,5'-Tetramethylbenzidine (TMB). Upon insertion into an infected canal, ConA selectively binds to polysaccharide components of the extracellular polymeric matrix of biofilms, resulting in a visible colour change from-colour-less-to-blue. This systcm cnablcs direct identification-of-biofilm presence independent of bacterial metabolic activity, making it particularly useful in detecting dormant or persistent infections. The invention provides a rapid, chairside diagnostic tool to enhance clinical decision-making prior to obturation.
