Abstract: The present invention is related to a bio derived cryo embedding medium which is intended for use in frozen section histopathology. The composition integrates hydroxypropyl cellulose, sodium alginate, glycerol, and sorbitol into a_gel based embedding medium that serves as an effective alternative to conventional Optimal Cutting Temperature (OCT) media. The formulation provides a clear to opalescent, water-soluble gel that offers sufficient mechanical support to biological tissues during cryostat sectioning, while simultaneously reducing ice crystal artefacts at sub-zero temperatures. By combining a plant derived polymer base with a natural polysaccharide matrix and cryoprotective plasticizers, the invention enhances tissue stability, cutting precision, and section integrity without interfering with routine histological staining. The use of glycerol and sorbitol improves flexibility and performance at low temperatures, thereby minimizing tissue distortion and sectioning defects. This bio compatible and biodegradable embedding medium reduces reliance on synthetic polymers, improves laboratory safety, and supports efficient workflow in diagnostic settings. The invention is particularly advantageous in diagnostic histopathology, surgical pathology, and margin assessment, where rapid and reliable frozen section analysis is essential.
