Abstract: This study presents the synthesis of novel Benzodiazocine analogues and their potential as potent antiviral agents. Benzodiazocines, featuring an eight-membered fused bicyclic core, adhere to Lipinski’s rule of five, ensuring drug-like properties. Structural modifications via Structure-Activity Relationship (SAR) enhanced their antiviral efficacy. In vitro assays, including cytotoxicity analysis and RT-qPCR-based studies, evaluated activity against SARS-CoV-2. Several analogues demonstrated promising antiviral potential, contributing to the ongoing pursuit of effective therapeutics against viral infections.
