Abstract: The present invention relates to an Ashwagandha phytoextract-loaded nanocarrier composition for management of alopecia and associated fungal infections. A standardized Withania somnifera phytoextract containing one or more marker withanolides is incorporated into a nanoscale delivery system selected from nanostructured lipid carriers, solid lipid nanoparticles, nanoemulsions, liposomes, phytosomes, polymeric nanoparticles and lipid-polymeric nanocarriers. The developed formulation demonstrated nanoscale particle characteristics, satisfactory extract loading, physicochemical compatibility, controlled and sustained release, enhanced skin permeation and improved follicular deposition of Ashwagandha phytoconstituents. The optimized formulation additionally demonstrated antioxidant, anti-inflammatory and antifungal activity against selected fungal organisms associated with scalp infections. The nanocarrier provides prolonged residence of the phytoextract at the scalp and facilitates delivery toward the hair follicular microenvironment. The formulation may further comprise a pharmaceutically acceptable antifungal agent and may be incorporated into a gel, lotion, serum, cream, shampoo or scalp spray. The invention provides a dual-action topical delivery platform intended to address hair loss and associated fungal or inflammatory scalp conditions while improving the pharmaceutical performance of Ashwagandha phytoextract.
