Abstract: ABSTRACT The present disclosure relates to apparatus and methods for simultaneous multi-sample genotype detection. The apparatus (100) includes a microfluidic cartridge (102) defining a plurality of reaction wells (104), each having a wettable interior surface (106) for gravity-driven sample distribution and a pre-loaded lyophilized reagent pellet (208) supporting isothermal nucleic acid amplification and colorimetric detection. A unified heating plate (108) with heating elements (110) and a temperature sensor (112) maintains all reaction wells (104) at a uniform isothermal amplification temperature through a single thermal control loop. A sealing assembly (116) employs a compliant sealing member (118) and linear actuator (120) to simultaneously seal all reaction wells (104) in a single actuation stroke. An optical capture assembly (122) captures all reaction wells (104) in a single image, enabling colorimetric genotype determination. The apparatus (100) provides simplified, rapid, multi-sample pharmacogenomic testing suitable for point-of-care environments. FIG. 1
