Abstract: ABSTRACT The present disclosure relates to electrochemical measurement of glucose concentration in single-use bioreactors. The apparatus (100) includes a housing (102) with a flow channel (104), an electrode substrate (106) positioned within the flow channel (104), and an enzyme layer (110) disposed over an electrochemical working electrode (108). The enzyme layer (110) contains glucose oxidase and restricts glucose transport to the electrochemical working electrode (108), ensuring glucose transport rate governs hydrogen peroxide generation rather than enzyme reaction kinetics. The enzyme layer (110) supplies hydrogen peroxide at a flux proportional to glucose concentration across a range of 0.1 to 20 grams per litre. The electrochemical working electrode (108) oxidizes the hydrogen peroxide and generates a current signal varying linearly with glucose concentration across the range. The resulting current signal enables real-time inline glucose monitoring during cell culture, providing accurate and continuous concentration measurements directly within the single-use bioreactor environment.
