Abstract: The present invention relates to novel apocynin-amine hybrid compounds of Formula (I), or pharmaceutically acceptable salts, solvates, hydrates, stereoisomers, tautomers, or prodrugs thereof, useful as acetylcholinesterase inhibitors. The invention further provides a process for preparation of the compounds comprising nucleophilic ring opening of an oxirane-substituted apocynin intermediate with selected secondary amines. The compounds comprise apocynin derivatives bearing pyrrolidine, piperidine, piperazine, benzylpiperidine, benzylpiperazine and related amino moieties. The disclosed compounds were evaluated for acetylcholinesterase inhibitory activity and subjected to molecular docking, density functional theory and molecular dynamics studies. Representative compounds ACT-1BP, ACT-4BP and ACT-NBA exhibited promising acetylcholinesterase inhibitory activity, supporting their potential utility as acetylcholinesterase inhibitor candidates.
