Abstract: Periodontitis is a prevalent chronic inflammatory disease characterized by progressive destruction of the periodontal ligament, alveolar bone, and surrounding soft tissues, which can ultimately result in tooth loss. The disease arises from a complex interaction between pathogenic bacterial biofilms and the host immune response, causing persistent inflammation and tissue breakdown. While conventional periodontal therapies focus on mechanical debridement to remove plaque and tartar, they often fail to fully control inflammation and bacterial recolonization, limiting long-term treatment success. Therefore, there is a critical need for adjunctive therapies that provide localized, sustained delivery of bioactive agents directly to diseased periodontal sites. Apigenin, a naturally occurring plant-derived flavonoid, is recognized for its potent anti inflammatory, antioxidant, antimicrobial, and tissue-regenerative properties. These multifunctional effects make apigenin an ideal candidate for addressing the pathological processes underlying periodontitis, including modulation of inflammatory pathways, inhibition of bacterial biofilms, and promotion of healing in periodontal tissues. This invention provides a novel local drug delivery system comprising an apigenin-containing gel formulated for administration into periodontal pockets and intrabony defects. The system is designed to achieve sustained and controlled release of apigenin at therapeutic levels, overcoming challenges such as salivary clearance, enzymatic degradation, and pH variability in the oral cavity. By maintaining effective drug concentrations at the site of periodontal destruction, the delivery system aids in reducing inflammation, controlling microbial growth, and supporting regeneration of bone and soft tissues. The localized nature of this delivery method offers significant clinical advantages, including targeted therapy with minimal systemic side effects, reduced dosing frequency, and enhanced patient compliance. It can be used as an adjunct to standard mechanical periodontal treatment to improve clinical outcomes such as pocket depth reduction, attachment gain, and bone regeneration in intrabony defects. The present invention specifically addresses the formulation and application of apigenin in a stable, bioavailable form within a biocompatible gel matrix suitable for periodontal use. This innovative drug delivery system leverages apigenin's multifaceted biological activities to provide a safe, natural, and effective adjunctive treatment for periodontitis. It represents a significant advancement in periodontal therapy by combining targeted local drug delivery with the therapeutic potential of a naturally derived bioactive compound to enhance healing and long-term periodontal health.
