Abstract: The present disclosure relates to a platform for antibiotic susceptibility testing (AST) of microbes and methods of fabricating and using the same. The platform comprises a substrate having a hydrophobic surface patterned with hydrophilic regions of varying sizes, and a polymeric layer having through-holes aligned with the hydrophilic regions. The hydrophilic regions are configured to retain predefined quantities of an antibiotic based on their size, thereby generating concentration gradients. An antibiotic solution is deposited and dried on the hydrophilic regions, followed by introduction of a microbial culture into the through-holes, and a viability indicator is added to detect microbial growth. A detectable change in the indicator is used to determine antibiotic susceptibility and minimum inhibitory concentration (MIC). The platform enables rapid, cost-effective, and multiplexed testing with reduced reagent consumption and assay time.
