Abstract: ANALYTICAL METHOD FOR QUANTITATIVE ESTIMATION OF 2-ACETYL-1-PYRROLINE IN INDIGENOUS AROMATIC RICE VARIETIES The present invention provides a cost-effective methodology for quantitative estimation of 2-acetyl-1-pyrroline (2AP), the principal aroma compound in small-grain aromatic rice. The method employs headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography–mass spectrometry (GC–MS/MS), wherein a pH-assisted protocol for volatilization is applied. Acid treatment establishes a “zero” reference level by eliminating detectable 2AP below odor threshold value (OTV) and limit of detection (LOD) by GC-MS, while alkali treatment enhances 2AP release, providing an amplified reference point. By comparing Peak areas and Total Ion Count (TIC) abundances of test samples against these defined baselines, the methodology enables reproducible and scalable estimation of aroma intensity even at very low concentrations. This inventive approach simplifies quantification, reduces cost and time, and provides a scientific basis for grading, quality assurance, and price-tagging of aromatic rice varieties for domestic and export markets.
