Abstract: The present invention relates to oral sustained-release pharmaceutical formulations of Origanum vulgare extract. Microbeads (160) comprise 3% w/v extract, 5% w/v sodium alginate and 0.05% w/v guar gum, and form by dropping a homogenized, dark-stirred, sonicated and de-bubbled mixture through a 21G needle into 5% w/v calcium chloride solution (150). The resulting calcium-alginate and guar-gum matrix is washed and air-dried or freeze-dried for three days. The microbeads constitute 40-90% w/w of an oral composition containing the defined diluent, disintegrant, glidant and lubricant ranges. The matrix remains intact under gastric conditions and undergoes greater swelling and release under intestinal pH conditions, thereby providing sustained release of the encapsulated extract. Hard gelatin capsules show controlled release over 480 minutes and retain assay, moisture and dissolution characteristics during six-month storage. Reference
