Abstract: ABSTRACT The present invention provides an intranasal Ramelteon drug delivery system (100) comprising a Ramelteon-loaded nanostructured lipid carrier (101), glyceryl monostearate as a solid lipid (102), oleic acid as a liquid lipid (103), Tween 80 as a surfactant (104), Poloxamer 407 as a stabilizer (105), an aqueous phase (106), an optimized NLC dispersion (107), and a Carbopol 934 mucoadhesive gel base (108). Ramelteon, the lipids and ethanol:acetone (1:1) are processed by solvent evaporation, stirring, probe sonication and cooling. A three-factor, three-level Box–Behnken design optimizes lipid concentration, surfactant concentration and sonication time. The optimized formulation provides 82.35% entrapment efficiency, 95.48 nm particle size and 92.37% cumulative release. The optimized NLC dispersion is incorporated into the Carbopol 934 gel base for intranasal delivery with modified release and nasal permeation characteristics.
