Abstract: [096] The present invention discloses an integrated next-generation sequencing system for microbial forensic attribution. An evidence collection assembly acquires an evidence sample and a synchronized field control under a common machine-readable identifier while recording environmental exposure. Evidence, field-control and reagent-control nucleic acids are converted into distinguishably indexed libraries and sequenced. A processing unit derives a contamination transfer matrix from control read sets, corrects microbial features of the evidence read set, normalizes the corrected features according to an environmental exposure index, and generates a multi- layer microbial forensic signature comprising taxonomic and strain-discriminating features. Mixed signatures are resolved into contributor components and compared with selected reference signatures to generate a calibrated association or exclusion output with uncertainty and quality-gate status. An evidence-package generator cryptographically binds the sample identifier, sequence data, processing parameters, reference version and output, thereby reducing contamination-induced false association, environment-induced false exclusion and post-analysis data alteration.
