Abstract: An integrated system and process for production of a biofuel from biomass is disclosed. Biomass feedstock is size-reduced and treated in a pretreatment reactor using a heat transfer fluid and/or a chemical reagent to produce pretreated biomass, subjected to enzymatic hydrolysis to produce a hydrolysate. The hydrolysate is separated into a fermentable sugar solution and a lignin-rich solid residue. The sugar solution is fermented with a microbial culture to produce a biofuel-containing broth, distilled to separate a biofuel product and a residual stream. The lignin-rich solid residue and residual stream are combusted in a byproduct combustion and heat recovery unit to generate heat, supplied to the pretreatment reactor through a heat transfer loop, while residual process water from distillation is recycled to the pretreatment or hydrolysis reactor through a water recycle loop. A controller regulates temperature, pressure, and agitation within the pretreatment reactor, reducing external energy and fresh water consumption. Reference Figure: FIG. 1
