Abstract: The present invention relates to an improved process for preparing and purifying N.meningitidis serogroup C polysaccharide-CRM197 protein conjugate with high conjugation efficiency and improved recovery, wherein the process uses optimized ratio of polysaccharide to CPPT to protein and optimized concentration of polysaccharide during conjugation. The conjugate obtained is further purified by concentration, diafiltering and maintaining a low transmembrane pressure (TMP) and applying back pressure. The method produces a conjugate with low free polysaccharide content.
