Abstract: An Improved Process for Preparation of a Natural Cytological Staining Composition Derived from Nyctanthes arbor-tristis The present invention discloses an improved process for preparation of a natural cytological staining composition derived from Nyctanthes arbor-tristis, comprising controlled drying of floral material at 35 to 70°C for 6 to 48 hours to obtain dried material having 5 to 12% moisture content, pulverization to a particle size of 100 to 500 µm, extraction using an aqueous alcoholic medium at a water to alcohol ratio of 20:80 to 80:20 (v/v) and a plant to solvent ratio of 1:5 to 1:20 (w/v) at 20 to 60°C for 30 to 240 minutes, followed by filtration, centrifugation at 3,000 to 8,000 rpm, pigment standardization to 10 to 60 mg/mL, optional incorporation of 0.05 to 5.0% (w/v) staining auxiliaries and storage at 2 to 30°C, thereby producing a stable natural staining composition exhibiting enhanced nuclear staining, improved chromatin differentiation, reduced background staining, improved extract clarity, high reproducibility and prolonged storage stability for cytological, histological and biological diagnostic applications.
