Abstract: The present invention relates to a glycoconjugate comprising polysaccharide derived from Streptococcus pneumoniae serotype 9N conjugated to a carrier protein and immunogenic compositions comprising said 9N glycoconjugate. The present invention also provides method for preparing said polysaccharide-protein conjugate based monovalent and multivalent immunogenic compositions and application of the immunogenic compositions for preventing S. pneumoniae infection or disease in a subject. The immunogenic composition of the present invention has advantages including mitigating epitope suppression while maintaining efficacy and antigenic integrity, cost-effective, has reduced levels of free polysaccharide and free proteins, improved purity profiles and improved immunogenicity.
