Abstract: The present disclosure relates to an electrochemical sensing platform for detection of nicotine in biological samples. The platform comprises an electrochemical sensing device (100) including a multilayer microfluidic cartridge structure (102) having a transparent PET layer, a first pressure-sensitive adhesive layer, a PMMA flow layer, a second pressure-sensitive adhesive layer, and a PMMA base layer defining a controlled fluid transport pathway. The device (100) further comprises a sample inlet (130), a sample pad (120a), a purification region (106), a sample-containing pad (120b), and a mini electrode chip (126). The mini electrode chip comprises a working electrode modified with a MXene-gold nanoparticle nanocomposite, a reference electrode, and a counter electrode, and is operatively coupled to a portable potentiostat (128) for electrochemical nicotine detection. The disclosure further provides methods for fabricating the sensing platform and detecting nicotine, enabling rapid, portable, sensitive, and point-of-care analysis.
