Abstract: The present invention relates to a herbal topical anti-inflammatory gel formulation comprising extracts obtained from leaves and seeds of Gloriosa superba for the management and treatment of pain, inflammation, swelling, and associated musculoskeletal disorders. The formulation was developed using Carbopol-940 as a gelling agent at a concentration of 1% w/w along with pharmaceutically acceptable excipients to obtain a stable and homogeneous gel suitable for topical administration. The anti-inflammatory activity of the developed herbal gel was evaluated using carrageenan-induced paw edema model in Wistar rats of either sex weighing about 160–180 g. The change in edema volume of rat hind paw was measured to assess therapeutic efficacy. The developed herbal formulation demonstrated significant topical anti- inflammatory activity and potentiation effect against induced inflammation. Further, the formulation was evaluated for physicochemical parameters including pH, viscosity, spreadability, extrudability, in vitro diffusion profile, and stability. The pH of the formulation was found to be approximately 6.8, which is compatible with normal skin pH. The formulation exhibited satisfactory stability under normal storage conditions and did not produce any signs of skin irritation. Stability studies were carried out in accordance with ICH guidelines, confirming the suitability and effectiveness of the herbal gel for topical anti-inflammatory application.
