Pulla Reddy College Of Pharmacy on September 16, 2026, for Ambrisentan Solubility Enhancement Using Liquisolid Method And Optimization With 32 Full Factorial Design.

Inventors include Dr. Latha Kukati; and Javeria Tamkeen.

The application for the patent was published on September 18, 2026, under issue no. 38/2026.

Abstract: ABSTRACT The present invention relates to a liquisolid compact system (100) and a method for enhancing the aqueous solubility and dissolution rate of ambrisentan, a poorly water-soluble BCS Class II drug. The system comprises ambrisentan dissolved in a non-volatile solvent to form a drug solution, admixed with a carrier material and a coating material to form a liquid-powder admixture, and further blended with a binder, glidant, and lubricant before compression into a compact. The amount of the non-volatile solvent and the carrier-to-coating ratio are optimized using a two-factor, three-level (3²) full factorial experimental design based on percentage drug release, angle of repose, and disintegration time as response variables. The optimized compact exhibits about a four-fold increase in aqueous solubility, an amorphous form of the drug, enhanced dissolution, and acceptable physicochemical stability under accelerated storage conditions.
