Abstract: ABSTRACT: Altering XynA Substrate Specificity in Bacillus amyloliquefaciens through Double Mutation in the Substrate Binding Site (SBS) This invention describes a bi-functional enzyme derived from Bacillus amyloliquefaciens xylanase A (XynA). A double mutant enzyme was developed by site-directed substitution of two conserved aromatic residues, tyrosine at positions 128 and 124, with alanine (Y128A/Y124A) within the substrate binding site. This targeted structural modification resulted in a non-obvious and synergistic enhancement of multifunctional activity, enabling efficient hydrolysis of both hemicellulosic and cellulosic substrates, including xylan, carboxymethyl cellulose, avicel, and filter paper. The engineered enzyme demonstrates extraordinary relative activity increases, including up to 1444% enhancement for avicel, while retaining usable xylanase activity and exhibiting thermostability under industrial conditions. The invention provides a cost-effective, eco- friendly alternative to enzyme cocktails and is particularly suitable for applications in biofuel production, pulp and paper processing, animal feed, textile, and food industries.
