Abstract: The present disclosure relates to a validated reverse-phase liquid chromatographic method for the determination of Treosulfan and its related impurities, and a process for development thereof. The method comprises chromatographic separation of Treosulfan using an optimized mobile phase and stationary phase combination, established through systematic design-based optimization of critical method parameters. The process further comprises subjecting Treosulfan to forced degradation under acidic, alkaline, oxidative, reductive, thermal, photolytic, and neutral conditions, followed by identification and structural characterization of the resulting degradation products using mass spectrometric techniques. The disclosed method exhibits stability-indicating capability, adequate resolution of Treosulfan from its degradation products and related impurities, and reproducibility suitable for regulatory validation. The invention further provides a computational toxicity assessment of the identified degradation products, offering utility for quality control, stability testing, and impurity profiling of Treosulfan formulations.
