Abstract: The present invention relates to a topical dual-drug film-forming pharmaceutical composition for promoting healing of diabetic and chronic wounds. The composition comprises Dimethyloxalylglycine dispersed within a polymeric film-forming hydrogel matrix and Amlodipine, or a pharmaceutically acceptable salt thereof, incorporated within Nanostructured Lipid Carriers comprising at least one solid lipid, at least one liquid lipid, and a surfactant. The Amlodipine-loaded Nanostructured Lipid Carriers are uniformly dispersed throughout the Dimethyloxalylglycine-containing hydrogel matrix. Upon topical application, the composition spreads over the wound and forms a flexible, adherent, breathable, and moisture-retentive film. The hydrogel matrix provides localized release of Dimethyloxalylglycine, while the Nanostructured Lipid Carriers provide sustained release of Amlodipine. The integrated dual-delivery system promotes angiogenesis, improves endothelial function, supports collagen deposition and re-epithelialization, and accelerates wound closure. The invention also relates to a process for preparing the composition by hydrogel formation, nanostructured lipid carrier preparation, and controlled incorporation of the nanocarrier dispersion into the hydrogel matrix.
