Abstract: ABSTRACT The present invention relates to a thiazole–pyrazine–thiadiazole amide derivative system (100) for anticancer applications. A pyrazine–thiadiazole intermediate formation module (101) couples 5-bromopyrazine-2-carboxylic acid with a thiadiazole boronic acid derivative. A carboxamide conversion module (102) forms a carboxamide, while a carbothioamide formation module (103) converts the carboxamide into a carbothioamide. A thiazole cyclization module (104) cyclizes the carbothioamide with 1,3-dichloropropan-2-one to form a thiazole intermediate. An azide-to-amine transformation module (105) converts the azide intermediate into an amine. A final amide coupling module (106) couples the amine with substituted aromatic carboxylic acids to form derivatives 13a–13j. A characterization module (107) establishes structural features, while an anticancer evaluation and molecular docking module (108) evaluates cytotoxic activity against MCF-7, A549, Colo-205 and A2780 cell lines and binding interaction with a kinase target.
