Abstract: [075] A thermosensitive in situ gel composition for sustained ocular drug delivery and a method for preparation thereof are disclosed. The composition comprises at least one therapeutically active pharmaceutical ingredient, one or more thermosensitive polymers configured to undergo sol-to-gel transition at ocular physiological temperature, at least one mucoadhesive polymer, one or more viscosity-modifying agents, buffering agents, and a pharmaceutically acceptable aqueous carrier. The composition remains in a liquid state during storage and administration and transforms into a gel upon contact with the ocular surface, thereby prolonging precorneal residence time and enabling sustained drug release. The disclosed method comprises dispersing thermosensitive polymers, incorporating mucoadhesive polymers and active pharmaceutical ingredients, adjusting formulation parameters, homogenizing, sterilizing, and packaging the formulation. The disclosed composition enhances ocular bioavailability, reduces dosing frequency, improves patient compliance, and minimizes rapid drug elimination associated with conventional ophthalmic dosage forms.
