Abstract: Title of the Invention: A thermo-responsive DJK-5 peptide-based sol-gel varnish composition and process for endodontic antibiofilm repair A thermo-responsive DJK-5 peptide-loaded sol-to-gel varnish composition and process for localized dental application are disclosed. The composition comprises DJK-5 peptide, Poloxamer F-127, HPMC K30, mannitol, glycerol, EDTA disodium, and purified water. The composition is maintained in a sol state under refrigerated conditions of about 2–8°C and is deliverable in the sol state to a dental or endodontic treatment site, where exposure to physiological temperature triggers in-situ sol-to-gel transformation to form an adherent gel or varnish layer. The formed layer provides localized sustained release of DJK-5 peptide and exhibits antimicrobial and antibiofilm activity, including against mature multispecies oral biofilms, with favorable cytocompatibility. The composition is useful for endodontic repair, regenerative dentistry, periodontal therapy, implantology, and other localized antimicrobial applications.
