Abstract: A targeted colon specific drug delivery system is disclosed. The system comprises a drug containing core, an intermediate modulation layer, a microbially responsive layer, and an outer enteric barrier. The enteric barrier protects the dosage form during gastric passage, while the microbial layer responds to colonic enzymatic conditions. The intermediate layer regulates water ingress and drug diffusion after activation. The system may be configured as a single unit or multiparticulate dosage form for controlled colonic availability of an active pharmaceutical ingredient.
