Abstract: The invention belongs to medicinal and pharmaceutical chemistry, particularly to indole-based Schiff base compounds, their preparation, and in vitro biological evaluation. Disclosed are compounds selected from (E)-2-(((4-(1H-indol-5-yl)phenyl)imino)methyl)phenol and (E)-1-(((4-(1H-indol-5- yl)phenyl)imino)methyl)naphthalen-2-ol, identified as INDP and INDN, respectively, prepared by condensing 5-(4-aminophenyl)-1H-indole with salicylaldehyde or 2- hydroxy-1-naphthaldehyde in the presence of para toluene sulphonic acid at 80 degree Celsius, followed by precipitation and recrystallization. Structural confirmation is performed using 1H NMR and 13C NMR analyses. The disclosure further provides in vitro methods for evaluating cellular response in HEK293, MCF7, and HeLa cells by overlaid montage imaging, MTT assay-based viability quantification, reactive oxygen species determination, and confocal microscopy, enabling assessment and comparison of cytotoxic and imaging-related response characteristics of INDP and INDN.
