Abstract: The present disclosure generally relates to a system for detecting EGFR gene mutations in cell-free DNA using a CRISPR-Cas12a-based diagnostic method is disclosed. The system comprises a guide RNA design module to create specific gRNAs targeting EGFR mutations, including exon 18 indels, exon 19 deletions, exon 20 insertions, and L858R and T790M point mutations. An amplification primer design module optimizes primers for Recombinase Polymerase Amplification (RPA) to amplify EGFR regions from cell-free DNA isolated via a sample preparation unit. The system integrates a reporter molecule labelled with a fluorophore and quencher to monitor Cas12a trans-cleavage activity. The CRISPR-Cas12a reaction unit processes the amplified product with Cas12a, gRNA, and the reporter molecule, enabling target-specific cleavage. A detection unit measures fluorescence intensity or visualizes test and control lines on a lateral flow strip. This system offers a rapid, sensitive, and precise diagnostic solution for EGFR mutation detection, aiding in personalized cancer therapy.
