Abstract: The present invention relates to a phantom-based stereotactic irradiation and imaging system and method for correlating radiation dose distribution with live-cell biological responses. The system comprises a tissue-mimicking phantom body defining a cavity for receiving a cell culture vessel, a spatial reference grid for defining coordinate positions, a stereotactic localization interface for imaging-based coordinate registration, and a radiation alignment interface for targeted irradiation. The method includes culturing cells in a glass-bottom vessel, embedding the vessel within an agarose-based phantom, performing cone-beam computed tomography (CBCT) for localization, generating a radiation plan assigning doses in the range of 0.5 Gy to 2.5 Gy to selected coordinates, delivering radiation using a stereotactic irradiation system, and performing live-cell confocal imaging at multiple time intervals. The invention enables spatially resolved mapping of radiation dose with cellular responses including nuclear morphology and cell-cycle progression.
