Abstract: The invention relates to computational drug discovery and cancer therapeutics, and more particularly to a system and method for identifying maleic acid anhydride derivatives as androgen receptor inhibitor candidates for prostate cancer treatment. A three-dimensional structure of an androgen receptor ligand-binding domain, including wild-type and/or mutant forms, is prepared and used to define a docking grid for computer-aided docking of a candidate library enriched in maleic acid anhydride derivatives including compounds A, B, and C and analogues thereof. Ranked candidates are filtered using predicted binding energies, docking scores, interaction profiles with key androgen receptor residues, and ADMET and drug-likeness characteristics, followed by biological evaluation in PC-3, DU145, and HEK- 293 cells using MTT and LDH release assays. Computational and biological outputs, including IC50 values, cytotoxicity profiles, and a selectivity index, are integrated, optionally as a composite score, to identify active and selective androgen receptor inhibitor candidates.
