Abstract: ABSTRACT A method, a composition, a system, and a kit for cancer stem cells (CSCs) enrichment for inhibitor screening and resistance therapy The present disclosure relates to a method, a composition, a system, and a kit for cancer stem cells (CSCs) enrichment for inhibitor screening and resistance therapy. Particularly, the method involves co-culturing the patient-derived OTSCC cell lines with patient-derived cancer-associated fibroblast (CAFs) cells from the same patient grown in a CAF-conditioned medium. The system and kit relate to investigation of the effect of pharmacological inhibitors on CSCs involved in tumor progression resistant to the therapy. The invention provides a CSC enriched stable cell model, obtained from the co-culturing stem cell and the cancer-associated fibroblasts (CAFs) to screen pharmacological inhibitors. The application is the screening the inhibitory potential of pharmacological inhibitors against CSCs in native environment in presence of CAFs.
