Abstract: ABSTRACT A SYNERGISTIC COMPOSITION LOADED IN NANOPARTICLES FOR MANAGEMENT OF GLIOBLASTOMA MULTIFORME AND PREPARATION PROCESS THEREOF Synergistic formulation consisting 2mg temozolomide, 2mg quercetin and 2mg resveratrol encapsulated in PLGA nanoparticles for effective management of glioblastoma multiforme is disclosed. Drug-loaded nanoparticles are fabricated using modified emulsification and solvent evaporation technique, incorporating atomization. The preparation method involves mixing 2ml PLGA solution in 6ml acetone, dissolving 2mg drug in DMSO, mixing drug with PLGA solution, feeding drug-PLGA solution to 10ml syringe, pumping it using syringe controller at 0.3ml/min constant flow rate, spraying it onto 1%(w/v) 200ml PVA solution placed on magnetic stirrer with 1500rpm speed while using ultrasonic atomizer at 130KHz frequency, evaporating residual acetone by placing nanoparticles on hot plate at 55oC for 20-30minutes, purification by ultracentrifugation at 80000xg for 30 minutes and washing. The PLGA-loaded antitumor drugs significantly downregulate expression of MMPs, miR-21 and various miRNAs, thereby effectively managing glioblastoma. Additionally, this formulation exhibits enhanced blood-brain barrier penetration, superior entrapment efficiency, and lower IC50 values.
