Abstract: Title: A Sustained-Release Mucoadhesive Lipid Nanoparticle Formulation for Intranasal Delivery. Amethod for preparing intranasal mucoadhesive drug formulation(as shown in fig 1) has been proposed in the present invention. The lipid nanoparticles dispersed within a chitosan-based polymer matrix for enhanced delivery of antidepressant drugs. The lipid nanoparticles are prepared using glyceryl behenate and polyethylene– polypropylene glycol, followed by purification and probe sonication to obtain a stable nanosized system. The nanoparticles are incorporated into a mucoadhesive matrix containing chitosan and glycerine to improve nasal retention and mucosal adhesion. The formulation provides enhanced ex vivo nasal permeation and prolonged nasal residence, high drug entrapment efficiency, and excellent colloidal stability. Furthermore, it enhances direct nose-to-brain transport, improves bioavailability, reduces systemic side effects, and offers an effective approach for the treatment of depression and related neurological disorders.
