Abstract: The present invention relates to a gastro-retentive floating tablet formulation of Stavudine for prolonged drug release and a process for its preparation. The formulation comprises Stavudine as the active pharmaceutical ingredient, one or more hydrophilic polymers for controlled drug release, one or more gas-generating agents to impart buoyancy, and pharmaceutically acceptable excipients. Upon contact with gastric fluid, the gas-generating agent produces carbon dioxide, which becomes entrapped within the hydrated polymer matrix, enabling the tablet to float in the stomach for an extended period. The floating system increases gastric residence time, provides sustained release of Stavudine over approximately 12–24 hours, minimizes fluctuations in plasma drug concentration, reduces dosing frequency, and enhances patient compliance. The invention further provides a simple, economical, and reproducible process for preparing the floating tablets by blending, granulation or direct compression, lubrication, and compression. The resulting tablets exhibit acceptable pharmaceutical characteristics, including uniform drug content, adequate hardness, low friability, rapid floating, prolonged buoyancy, and controlled drug release, making them suitable for the effective oral delivery of Stavudine.
