Abstract: The present invention discloses a phytoflavonoid nano-in-hydrogel topical drug delivery system comprising solid lipid nanoparticles (SLNs) co-encapsulating quercetin (100 mg ) and apigenin (100 mg ) within a glyceryl monostearate lipid core (5.0 g), stabilized by Poloxamer 188 (1.75 g ), and dispersed in a 1.0% Carbopol 940 hydrogel (pH 5.82), for targeted topical treatment of atopic dermatitis (AD). The therapeutic rationale is supported by in-silico molecular docking against the IL-3 receptor (1IAR), where apigenin and quercetin showed binding affinities of -7.49 kcal/mol and -7.37 kcal/mol respectively, indicating potent inhibition of immune cell activation. The optimized formulation, prepared by hot high-shear homogenization (15000 rpm ) and probe ultrasonication (10 min ), achieves a particle size of 178.6 nm , PDI of 0.242 , and zeta potential of -28.4 mV . Quercetin inhibits NF- B/MAPK signaling and suppresses cytokines (IL-4, IL-13, TNF- ), while apigenin blocks the master switch TSLP/JAK2- STAT3 and upregulates barrier proteins, providing synergistic coverage of AD pathophysiology.
