Abstract: Title of Invention A Simplified Synthetic Process for Preparing Salcaprozic Acid and Salts Thereof for Oral Drug Delivery Abstract The present invention provides a simplified process for preparing salcaprozic acid, being 8-(2-hydroxybenzamido)octanoic acid, and pharmaceutically acceptable salts thereof, including sodium salcaprozate (SNAC) and potassium salcaprozate. Salicylamide is cyclised with carbonyldiimidazole to 2H-benzo[e][1,3]oxazine-2,4(3H)-dione, which is N-alkylated with an ethyl 8-halooctanoate in the presence of an inorganic carbonate base and a catalytic alkali-metal iodide, preferably potassium iodide, to give the corresponding alkylated benzoxazinedione. Aqueous-base hydrolysis then simultaneously cleaves the ester and opens the ring to afford salcaprozic acid, which is optionally converted to the sodium or potassium salt and isolated by antisolvent precipitation. The process is operated without special colour-removing reagents and without an inert-gas atmosphere, and affords the products as white-to-off-white solids of at least 99% HPLC purity, suitable for oral drug-delivery compositions.
