Abstract: A sequential microbiota-activated enzyme-responsive oral delivery composition for site-specific release of biologic therapeutics is disclosed. The composition comprises a biologic therapeutic agent encapsulated within a nanoparticulate carrier, a protease-modulating intermediate layer surrounding the nanoparticulate carrier, a secondary enzyme-responsive barrier layer, a first polysaccharide layer degradable by gut microbiota-associated enzymes, and an enteric outer coating resistant to gastric degradation. Upon oral administration, the enteric outer coating protects the composition in gastric conditions, while degradation of the first polysaccharide layer by microbiota-derived enzymes within a target intestinal region exposes the secondary enzyme-responsive barrier layer. Sequential release of the protease-modulating intermediate layer occurs prior to release of the biologic therapeutic agent, thereby enhancing localized protection and site-specific intestinal delivery of biologic therapeutics including peptides, proteins, antibodies, hormones, enzymes, and nucleic acid therapeutics.
