Abstract: Title: A Scalable Bioprocess for Sustainable Production of Citramalic Acid. The present invention relates to a process for producing citramalic acid using a genetically modified yeast strain and food waste hydrolysate-derived carbon sources. The process comprises introducing an exogenous codon-optimized citramalate synthase gene, a mitochondrial tricarboxylate transporter gene, and a citramalate export transporter gene into a yeast strain to enhance citramalate biosynthesis and secretion. The genetically modified yeast strain is cultivated in a fermentation medium comprising food waste hydrolysate-derived glucose, nitrogen sources, mineral salts, trace elements, and optional secondary carbon sources in a batch bioreactor under optimized fermentation conditions. The citramalate synthase catalyses the formation of citramalate from pyruvate and acetyl-CoA, while the transporter proteins improve intracellular carbon flux and extracellular citramalate export. Medium composition and process parameters are optimized using plackett-burman design, central composite design, artificial neural network modelling, and genetic algorithm optimization. The process provides a sustainable and economically viable platform for producing citramalic acid as a renewable precursor for methyl methacrylate, polymethyl methacrylate, and other industrial chemicals.
