Inventors include Dr. Mudasir Gani; Shabnum Mumtaz; Baseerat Ul Ann; Pawar Shubham Tarasing; Sahil Ahmad Dar; Shivani; Rajesh Kumar Jena; and Divyanshi Kashyap.
The application for the patent was published on September 11, 2026, under issue no. 37/2026.
Abstract: This invention describes a rapid extraction-free polymerase chain reaction (PCR)-based protocol for detecting Bombyx mori bidensovirus (BmBDV) directly from infected silkworm biological samples. The protocol replaces conventional multi-step nucleic acid extraction with a controlled alkaline pretreatment followed by neutralization to generate PCR-compatible templates. A biological sample is treated with sodium carbonate solution, diluted with water, incubated under predetermined conditions to promote disruption of viral particles and release of viral nucleic acid, and subsequently neutralized using hydrochloric acid prior to direct PCR amplification. Detection is performed using primers targeting a conserved region of the viral VD1 gene to generate a diagnostic amplicon of approximately 274 base pairs. The protocol substantially reduces processing time, reagent consumption, laboratory handling, contamination risk, and nucleic acid loss while maintaining reliable amplification efficiency. The invention is suitable for routine diagnostics, high-throughput screening, epidemiological surveillance, and disease management applications in sericulture.
