Abstract: The present disclosure proposes pharmaceutical analysis and provides a stability-indicating reverse-phase high-performance liquid chromatographic (RP-HPLC) method for determination, separation, and characterization of Acoltremon and its degradation products. Acoltremon or pharmaceutical compositions containing Acoltremon are subjected to forced degradation under acidic, alkaline, oxidative, reductive, thermal, hydrolytic, and photolytic stress conditions to obtain degradation products. The stressed samples are separated using an RP-HPLC system equipped with a C18 column under isocratic conditions employing an acetonitrile and trifluoroacetic acid buffer mobile phase with detection at about 223 nm. The chromatographically resolved components are analyzed using photodiode array detection and further characterized by liquid chromatography–tandem mass spectrometry (LC–MS/MS) to determine mass-to-charge ratios and fragmentation patterns for structural elucidation of impurities. The validated method is rapid, precise, robust, and suitable for impurity profiling, stability testing, and routine quality control of Acoltremon pharmaceutical formulations.
