Abstract: Title of the Invention: A quinazoline derivative compound for breast cancer treatment and synthesis process of the compound thereof A quinazoline derivative compound for breast cancer treatment and synthesis process of the compound thereof are disclosed for targeting HER2-mediated signaling in breast cancer. The compound is synthesized via a simplified process involving N-(4-methoxyphenyl)-2-phenylacetamide and benzonitrile. The process comprising the steps of: reacting N- (4-methoxyphenyl)-2-phenylacetamide with benzonitrile in the presence of 2-chloropyridine and trifluoromethanesulfonic anhydride in a solvent, dichloromethane to form a mixture; heating the reaction mixture at 45°C for 16 hours to obtain an intermediate, 2-benzyl-7-methoxy-4-phenylquinazoline, dissolving the intermediate in ethanol and reacting with 3-fluoro benzaldehyde, refluxing the solution at 110°C for 1 hour and 55 minutes and filtering, washing, and recrystallizing the final product from boiling ethanol to obtain the quinazoline derivative compound. The final compound demonstrates high selectivity and cytotoxicity toward MCF-7 cells with minimal effects on normal cells. Its improved pharmacokinetic properties and synthesis route position it as a promising anti-breast cancer therapeutic.
