Abstract: TITLE: A PROCESS OF PREPARATION OF PLATELET-RICH FIBRIN– DERIVED CONDITIONED MEDIA TO ENHANCE PERIODONTAL LIGAMENT REGENERATION APPLICANT: 1. MEENAKSHI ACADEMY OF HIGHER EDUCATION AND RESEARCH 2. MEENAKSHI AMMAL DENTAL COLLEGE AND HOSPITAL ABSTRACT The present invention discloses a process for preparation of platelet-rich fibrin (PRF)–derived conditioned media intended for periodontal ligament regeneration. The process includes isolating plasma albumin from autologous whole blood by centrifugation and collecting the supernatant, followed by denaturation through controlled heating to obtain denatured plasma albumin. Advanced Platelet-Rich Fibrin Plus (A-PRF+) is prepared by centrifugation of autologous blood to obtain a fibrin-rich pellet, while Liquid PRF (i-PRF) is obtained as a supernatant fraction through low-speed centrifugation. Albumin-PRF (Alb- PRF) is formed by combining liquid PRF with denatured plasma albumin. The A-PRF+ and Alb-PRF are subsequently mixed with serum-free Dulbecco’s Modified Eagle Medium to form a bioactive mixture. The mixture is incubated under controlled conditions, followed by centrifugation and sterile filtration to obtain a conditioned media, under aseptic conditions to obtain platelet-rich fibrin (PRF)–derived conditioned media
