Abstract: Title: A PROCESS FOR THE SYNTHESIS OF INDOLE-3-CARBOXALDEHYDE DERIVATIVES AND TRIS(INDOLYL)METHANE DERIVATIVES USING METHANOL. A method for the synthesis of tris(indolyl)methane derivatives (4a–k) as shown in fig 2 through potassium persulfate-mediated oxidative C–H functionalization of indoles is disclosed. The process involves treating an indole compound with methanol in the presence of potassium persulfate as the sole oxidant at 80°C for 5 h, affording 3,3'-((3H-indol-3-ylidene) methylene) bis(1H-indole) intermediates (3a–k) in high yield. Control studies demonstrate that the transformation proceeds efficiently without the requirement for transition-metal catalysts or bases. Potassium persulfate exhibits superior performance compared with other oxidants, including hydrogen peroxide, tert-butyl hydroperoxide, and di-tert-butyl peroxide. The resulting intermediates are reduced using sodium borohydride in ethanol at room temperature to furnish tris(indolyl)methane derivatives (4a–k) in good to excellent yields. The method features readily available reagents, mild reaction conditions, operational simplicity, short reaction time, and broad substrate scope, providing an economical, scalable, and environmentally friendly approach for the preparation of biologically important tris(indolyl)methane compounds.
