Abstract: The present invention relates to a process for the preparation of an immunomodulatory extract from Berberis vulgaris and its evaluation using in-vitro and in-vivo experimental models. The process comprises collection and authentication of plant material, followed by drying, pulverization, and extraction using aqueous and alcoholic solvents to obtain bioactive constituents. The prepared extracts are subjected to phytochemical screening and evaluated for immunomodulatory activity. The in-vitro evaluation includes neutrophil adhesion test indicating enhancement of non-specific immune response. The in-vivo evaluation comprises delayed-type hypersensitivity (DTH) response and immunoprophylactic studies in experimental animals. The extracts exhibit significant immunostimulatory activity by enhancing neutrophil adhesion and modulating cellular immune response. Further, the extracts demonstrate protective effect against Escherichia coli-induced infection by reducing mortality in treated animals as compared to control. The present invention thus provides a standardized process for the preparation of a plant-based immunomodulatory extract with significant therapeutic potential, offering a safe, effective, and cost-efficient alternative to synthetic immunomodulatory agents.
