Abstract: ABSTRACT A PROCESS FOR THE AQUEOUS CATALYTIC SYNTHESIS OF [1,2,4]TRIAZOLO[4,3-a]PYRIDIN-3(2H)-ONE FROM SEMICARBAZIDE AND 2-CHLOROPYRIDINE The invention relates to an organic synthetic process for preparing [1,2,4]triazolo[4,3-a]pyridin-3(2H)-one, wherein semicarbazide or semicarbazide hydrochloride is directly cyclocondensed with 2- chloropyridine in water as the sole reaction medium in the presence of catalytic p-toluenesulfonic acid, preferably 1-10 mol%, more preferably about 5 mol%. The reaction is conducted under heating, preferably at 120 °C to 150 °C, more preferably about 130-140 °C, under reflux conditions, using approximately one equivalent of semicarbazide relative to 2-chloropyridine. The process affords [1,2,4]triazolo[4,3-a]pyridin-3(2H)-one in isolated yield greater than 78%, without chromatographic purification, and is scalable to at least 10 g batch scale without substantial modification of reaction conditions. The invention further encompasses the product obtained by the process. Dated this 29th day of May, 2026 (Suvarna Pandey) Regd. Patent Agent [IN/PA-1592] Of RNA, IP Attorneys Agent for the Applicant
