Abstract: The present invention discloses a novel process for the synthesis, purification, and characterization of cyclic di-guanosine monophosphate (c-di-GMP) analogs functionalized with small photocrosslinkers of photoreactive tags and affinity tags by introducing a new synthetic approach combining phosphoramidite and H- phosphonate chemistry, an efficient and modular preparation of minimally modified cyclic di-guanosine monophosphate (c-di-GMP) analogs. The synthesized functionalized c-di-GMP analogs presented in four general structural formulae (Formula 1 to Formula 4) comprising an O6-functionalized guanosine residues, an AT group representing affinity tag, a PT group representing photoreactive tag which can diazirine or benzophenone or diazo or aryl azide, phosphodiester linkages and linkers that are alkyl or substituted alkyl chains bonded with heteroatoms. The analogs are designed for use in photoaffinity labelling (PAL) and chemoproteomics to investigate the interactome of c-di-GMP, a key bacterial second messenger involved in regulating various physiological and pathogenic processes such as biofilm formation, motility and virulence..
