Abstract: ABSTRACT The present invention relates to a process (100) for synthesis and biological evaluation of amide derivatives of naphthyridines as anticancer agents. The process comprises synthesis (102), preparation of derivatives (104), characterization (106), biological activity evaluation (108), molecular docking (110), and structure- activity relationship analysis (112). The synthesis involves sequential chemical transformation of an aminopyridine derivative to obtain a naphthyridine intermediate, followed by cyclization, chlorination, palladium-catalysed coupling with a 1,3,4-oxadiazole-containing boronic acid derivative, reduction of a nitro group, and coupling with aromatic carboxylic acids to obtain the amide derivatives. The synthesized derivatives are characterized using ^1H NMR spectroscopy, ^13C NMR spectroscopy and mass spectrometry. The biological activity is evaluated against MCF-7, A549, Colo-205 and A2780 human cancer cell lines using an MTT assay. Molecular docking against EGFR tyrosine kinase and structure-activity relationship analysis are further performed to identify potent anticancer derivatives.
