Abstract: The present invention relates to a process for preparing a modified-release oral pharmaceutical tablet comprising 650 mg of doxofylline per unit dosage form. The process comprises separately conditioning hydroxypropyl methylcellulose K100M (112 to 118 mg) and hydroxypropyl methylcellulose K4M (63 to 67 mg) at 38 to 45°C and 40 to 45% relative humidity for 4–8 hours, followed by sequential polymer matrix formation, high-shear granulation using a binder solution of polyvinylpyrrolidone K30 in isopropyl alcohol, fluid-bed drying at 48–55°C to obtain granules having 2.0 to 3.0% (w/w) residual moisture, milling to a median particle size (D50) of 220 to 300 µm, polymer relaxation at 42 to 48% relative humidity for 8 to 16 hours, lubrication, and compression at 15 to 19 kN to produce a modified- release tablet having reproducible mechanical properties and dissolution characteristics through a process-engineered hydrophilic polymer matrix.(Figure 1)
