Abstract: ABSTRACT A PROCESS FOR PREPARATION OF A FERROCENE-COUMARIN COMPOUND The present disclosure provides a one-pot multicomponent process for preparing a coumarin-based compound. The process comprises dissolving 4-hydroxycoumarin, malononitrile, and a piperazine catalyst in a methanol: water solvent system at room temperature; adding ferrocene carboxaldehyde to the dissolved mixture; heating the reaction mixture under reflux at about 80°C for about 30 minutes to form a precipitate; collecting the precipitate by filtration; washing the precipitate with methanol: water; and recrystallizing from methanol: dichloromethane to obtain the compound. The compound prepared by the process inhibits aggregation of intrinsically disordered proteins, particularly TDP-43A315T. [Figure 1]
